I don't have access to 64-bit Windows to try this unfortunately, and I
have no issues running on a 32-bit Windows Server 2003 and Windows XP
Pro (started with Buildbot 0.7.7 and Twisted 2.1.0; now it's 0.7.8 and
8.1.0, respectively).

I'd go up the chain and check if there are any issues on this platform
with Twisted and Python. Also, I'd suggest installing the latest version
of Python, 2.5.2, and trying the 64-bit build first (they have both
x86_64 and Itanium builds). I don't know if Python team runs tests with
32-bit version on 64-bit Windows, but I'm sure they tested 64-bit build
on the matching platform and you can be sure it works (so you only have
Twisted and Buildbot to troubleshoot). I checked Twisted and Buildbot
masters and they don't seem to have any 64-bit Windows slaves:
